Introduction

Macrophage migration inhibitory factor (MIF or MMIF) also known as glycosylation-inhibiting factor (GIF), L-dopachrome isomerase, or phenylpyruvate tautomerase is a protein that in humans is encoded by the MIF gene. Macrophage migration inhibitory factor assembles into a trimer composed of three identical subunits. Each of these monomers contain two antiparallel alpha helices and a four-stranded beta sheet. The monomers surround a central channel with 3-fold rotational symmetry. MIF plays a role in the regulation of macrophage function in host defense through the suppression of anti-inflammatory effects of glucocorticoids. It is an inflammatory mediator associated with rheumatoid arthritis (RA) severity. Additionally, evidence suggests that there is a correlation between MIF production and metastatic potential in colorectal cancer.


Principle of the Assay  

This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-MIF polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-MIF polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were  used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the MIF amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of MIF can be calculated.
