Introduction

  1. Cartilage is a resilient and smooth elastic tissue, rubber-like padding that covers and protects the ends of long bones at the joints, and is a structural component of the rib cage, the ear, the nose, the bronchial tubes, the intervertebral discs, and many other body components. It is not as hard and rigid as bone, but it is much stiffer and much less flexible than muscle.
  2. Tolonium chloride (INN, also known as toluidine blue or TBO) is a blue cationic (basic) dye used in histology and sometimes clinically. Toluidine blue solution is used in testing for lignin, a complex organic molecule that bonds to cellulose fibres and strengthens and hardens the cell walls in plants. A positive toluidine blue test causes the solution to turn from blue to pink. The dye is sometimes used by surgeons to help highlight areas of mucosal dysplasia (which preferentially take up the dye compared to normal tissue) in premalignant lesions (e.g. leukoplakia).

Protocol (Only for a reference)

  1. Conventional decalcify, embedding, fix.
  2. Add paraffin section into xylene twice.
  3. Wash series of alcohol each for 1min by tap water.
  4. Dip dyeing in the cartilage staining solution. The dyeing time is wholly different by different tissue.
  5. Wash for 2 mins by tap water, blot moisture by filter paper.
  6. Acetone differentiates to the degree of cartilage cells is bluish violet and Clearly visible.
  7. Dehydrated ethanol, dimethylbenzene transparent, neutral gum seal.

Dyeing results:

Cartilaginous, osteogenic cells are red violet; The background is light blue.


Note

1. For the dyeing of tissue that is more difficult to tint, the dip dyeing time should be extended.
