Description

TRIpure Reagent is a broad-spectrum total RNA extraction reagent. It is fast and easy to use, with bright colours and easy stratification. The reagent is used to extract total RNA from animal tissues, plant material, various microorganisms and cultured cells. The method provides good separation of small amounts of tissue (50-100mg) and cells (5 x 10⁶) as well as large amounts of tissue (≥1g) and cells (>10⁷). The samples were sufficiently lysed in TRIpure while being able to maximise RNA integrity. After centrifugation with chloroform, the solution is divided into three layers: an upper colourless aqueous phase, an intermediate layer and a lower red organic layer, with the RNA distributed in the supernatant layer. The extracted total RNA is free from protein and DNA contamination and can be used in a variety of routine molecular biology experiments such as RT-PCR, Real-time RT-PCR, Northern blot, Dot Blot, in vitro translation, etc. The RNA is then collected and precipitated in isopropanol to obtain total RNA.

TRIpure reagent facilitate the precipitation of a wide range of RNAs of different molecular weights and sizes from different species. For example, agarose gel electrophoresis of RNA extracted from rat liver and stained with ethidium bromide revealed many discrete high molecular weight bands between 7 kb and 15 kb (mRNA and hnRNA components), two dominant ribosomes of ~5 kb (28S) and ~2 kb (18S), and low molecular weight RNA between 0.1 and 0.3 kb (tRNA, 5S). When the extracted RNA is diluted with TE the A260/A280 ratio is ≥1.8. Note that with normal agarose gel electrophoresis, the 28S position is approximately 2 kb and the 18S position is approximately 1 kb, and the position varies considerably with different gel concentrations.
