Description
This kit uses an improved SDS-alkaline lysis method to lyse cells and specifically digests the yeast cell wall with a wall-breaking enzyme, and can isolate high-purity plasmid DNA from yeast culture broth within 1 hour. After the yeast is collected, the wall-breaking enzyme is added to remove the cell wall, and then the cells are lysed by alkaline fission. The silicon matrix membrane in the centrifugal adsorption column selectively binds the plasmid DNA in the solution under the condition of high salt and low pH value, and then deproteinizes the cells. A low-salt, high-pH elution buffer removes impurities and other bacterial components, followed by elution of pure plasmid DNA from the silica-based membrane.
