Description:
The enhanced miRNA first-strand synthesis kit adopts the principle of Poly(A) tailing method, firstly add a Poly(A) tail to the 3'end of the miRNA, and then use Anchored oligo(dT)-universal tag universal reverse transcription primers for reverse transcription reaction. Finally, the first strand of cDNA corresponding to the miRNA is generated.
Note:The kit must be used in conjunction with the enhanced miRNA fluorescence quantitative detection kit (BPC1054)
- Steps
Poly (A) tailing and reverse transcription reaction at the 3’ end of miRNA (first strand synthesis)
1. Thaw 2 × miRNA RT Reaction Mix and mix well. Put the RT Enzyme Mix on ice for later use. Add the following reagents to a total volume of 20μl (add miRNA RT Enzyme Mix at the end).

Note: miRNA RT Enzyme Mix is very viscous, and the solution is easily adsorbed on the tube wall and outside of the pipette tip to cause loss. Please spin off the tube before use, and avoid adhesion loss on the outer wall of the pipette tip.
*The total RNA used in the reaction must contain small molecules RNA (miRNA)
2. Gently mix the above-prepared reaction solution with a pipette, centrifuge briefly and react at 42°C for 60 minutes.
3. Heat at 85°C for 5 seconds to inactivate miRNA RT Enzyme Mix.
