Introduction 

Insulin-like growth factor 1 (IGF-1), also called somatomedin C, is a member of the insulin family of polypeptide growth factors.It is also one of three protein hormones that share structural similarity to insulin. IGF-1 has a molecular weight of 17,066 daltons, consists of 70 amino acids in a single chain with three intramolecular disulfide bridges. It is produced primarily by the liver as an endocrine hormone as well as in target tissues in a paracrine/autocrine fashion. Its primary action is mediated by binding to its specific receptor, the Insulin-like growth factor 1 receptor, initiates intrIGF-1llular signaling. It is a primary mediator of the effects of growth hormone (GH). It also plays an important role in childhood growth and continues to have anabolic effects in adults.


Principle of the Assay  

This kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. The purified anti- IGF-1 antibody was pre-coated onto 96-well plates. And the HRP conjugated anti- IGF-1 antibody was used as detection antibodies. The standards, test samples and HRP conjugated detection antibody were added to the wells subsequently, mixed and incubated, then, unbound conjugates were washed away with wash buffer. TMB substrates (A & B) were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the IGF-1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of IGF-1 can be calculated.
