Introduction

Interleukin-23 subunit alpha(IL-23) is a 189-amino acid protein that in humans is encoded by the IL23A gene. It is composed of the IL12 p40 subunit and the p19 subunit (IL23A), and is produced by dendritic cells and macrophages. Moreover, IL-23 is stimulated by Danger Signals, including cell debris, and directs memory T cells to Th17 response. It is an important part of the inflammatory response against infection. Recently, IL-23 has been implicated in the development of cancerous tumors. Langowski et al. (2006) showed that IL23 is an important molecular link between tumor-promoter proinflammatory processes and the failure of the adaptive immune surveillance to infiltrate tumors.


Principle of the Assay  

This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- IL-23 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- IL-23 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the IL-23 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of IL-23 can be calculated.
