Introduction
Glucagon-like peptide-1 (GLP-1) is an incretin derived from the transcription product of the proglucagon gene. The major source of GLP-1 in the body is the intestinal L cell that secretes GLP-1 as a gut hormone. It is an intestinal incretin that regulates glucose homeostasis through stimulation of insulin secretion from pancreatic beta cells and inhibits appetite by acting on the brain. Thus, it is a promising therapeutic agent for the treatment of type-2 diabetes mellitus and obesity. Hwang Ji et al study presents a possible macro- (genome level) and micro- (gene/exon level) evolution mechanisms of GLP1 and GLP1R, which allows them to acquire selective interactions between this ligand-receptor pair. This information may provide critical insight for the development of potent therapeutic agents targeting GLP1R. Compound K (CK) is a major metabolite of ginsenosides that is absorbed. It is a potential agent that has multiple biological functions in the body via GLP-1 secretion and TGR5 activation.
Principle of the Assay
This kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. The purified anti- GLP-1 antibody was pre-coated onto 96-well plates. And the HRP conjugated anti- GLP-1 antibody was used as detection antibodies. The standards, test samples and HRP conjugated detection antibody were added to the wells subsequently, mixed and incubated, then, unbound conjugates were washed away with wash buffer. TMB substrates (A & B) were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the GLP-1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of GLP-1 can be calculated.
